I use high-throughput magnetic tweezers to monitor RNA replication in real time at the single-molecule level, revealing how individual polymerase complexes synthesize RNA under controlled mechanical force. This approach enables quantitative analysis of replication kinetics, pausing, and error formation across thousands of parallel reactions.
